Selective correspondence between NFκB-associated chromatin accessibility and transcription in matched AML specimens

This record presents a secondary matched RNA–ATAC analysis of acute myeloid leukaemia (AML) specimens from data associated with Ochi et al., Chromatin landscape and epigenetic heterogeneity of acute myeloid leukaemia. The analysis tests whether NFκB-associated chromatin accessibility corresponds selectively to NFκB-related transcriptional output relative to prespecified GR and p53 regulatory axes. Across 1,366 matched AML RNA–ATAC specimens, NFκB-associated accessibility showed a strong positive association with NFκB-related transcriptional output after adjustment for cohort and biological composition (HC3-adjusted β = 0.445, 95% CI 0.403–0.487, P = 3.16 × 10⁻83). The association was concordant in Kyoto and Sweden and exceeded the corresponding NFκB RNA cross-axis associations. Neither GR nor p53 demonstrated comparable stable same-axis correspondence. The deposited v1 package contains the working manuscript, Figures 1–3, figure legends, aggregate/model-level statistical outputs, and reproducibility/provenance material. Specimen-level joined RNA–ATAC tables, sample mappings, and other specimen-level derived outputs are intentionally excluded from the public package. The findings support selective cross-modal NFκB accessibility–transcription correspondence in AML. They do not establish causal transcription-factor activity, direct NFκB occupancy, enhancer-to-gene regulation, temporal directionality, or TP53 alteration as the upstream cause of the observed NFκB-associated state. Abstract

The association was concordant in Kyoto and Sweden and exceeded the corresponding NFκB RNA cross-axis associations. Neither GR nor p53 demonstrated comparable stable same-axis correspondence. The deposited v1 package contains the working manuscript, Figures 1–3, figure legends, aggregate/model-level statistical outputs, and reproducibility/provenance material.
