Molecular Architecture, Synthetic Process Optimization, Peer Mechanistic Defense, and In Silico Modeling of an Ultra-Short-Acting β1-Blocker (C23H31NO6S)
Targeted cardioselective interventions during acute hemodynamic emergencies demand ultra-short elimination kinetics and strict peripheral restriction. Herein, we structurally resolve a novel candidate soft-drug entity with the stoichiometric formula C23H31NO6S, chemically designated as 2-(methylsulfonyl)ethyl (2E)-3-(4-(3-(tert-butylamino)-2-hydroxypropoxy)naphthalen-1-yl)prop-2-enoate. The scaffold couples an aryloxypropanolamine pharmacophore to a metabolically labile (methylsulfonyl)ethyl ester engineered for rapid systemic degradation (t1/2 ≈ 5–7 minutes). We present a chemoselectively re-engineered four-stage synthetic continuum that eliminates late-stage chiral attrition via enantiopure (2S)-epichlorohydrin and replaces hazardous high-temperature Lewis acid transesterification with mild biocatalytic transesterification (immobilized Candida antarctica lipase B, Novozym 435 at 42°C), completely suppressing thermal acrylate polymerization. Stereochemical integrity is established through CSP-HPLC (ee > 99.4%) and optical rotation parameters ([α]D25 = -24.6°). Crucially, this work directly addresses deep physiological and mechanistic boundaries: differentiating erythrocyte cytosolic esterases (Esterase D) from hepatic hCES1, evaluating in silico cardioselectivity against the Landiolol benchmark, investigating glutathione S-transferase (GST) nucleophilic catalysis risks, and defining industrial turnover constraints. Finally, an executable single-compartment parent-metabolite pharmacokinetic model confirms titratable steady-state IV micro-infusion dynamics and rapid bi-phasic renal elimination, providing an actionable theoretical and computational baseline for wet-lab institutional translation.
