Location, Location, Location: Membranous Nectin-4 as a Biomarker for Enfortumab Vedotin
Combination therapy with enfortumab vedotin, a Nectin-4-targeted antibody-drug conjugate, and pembrolizumab, an anti-PD1 antibody, recently supplanted platinum-based chemotherapy as standard first-line systemic therapy for patients with muscle-invasive or metastatic urothelial cancer. A composite biomarker integrating membranous Nectin-4 expression and NECTIN4 gene amplification is predictive of response to enfortumab vedotin.
