Associative relationship of the T704C polymorphic locus of the AGT gene with the development of circulatory system diseases in miners of the South Kuzbass agglomeration with occupational lung dust pathology
Introduction. The work activities of employees in the coal mining industry are associated with the combined exposure to harmful and hazardous production factors, which contribute to the development of not only occupational but also work related pathology, in particular diseases of the circulatory system. The key mechanism of the development of multifactorial diseases, including arterial hypertension and ischemic heart disease, is the dysfunction of the renin-angiotensin-aldosterone system, the activity of which is genetically determined. The study aims to analyze the contribution of the polymorphic T704C variant of the gene of the renin-angiotensin-aldosterone system to the development of circulatory system diseases in miners of the South of Kuzbass with occupational lung dust pathology.Materials and methods. The authors conducted an examination of 122 trainee workers employed in key professions at mines in the south of Kuzbass: 75 miners with a previously established diagnosis of "occupational lung pathology" complicated by diseases of the circulatory system (the main group), and 47 miners with the same occupational diagnosis but without concomitant cardiovascular pathology (the comparison group). All patients underwent Real-Time PCR genotyping of polymorphic variants of the AGT (T704C) gene using TaqMan probes.Results. In a study of the polymorphic locus of the AGT gene (T704C), it was found that carriage of the mutant C allele (OR=1.71; 95% CI (1.003–2.9); p=0.048) and the heterozygous T/C genotype (OR=2.5; 95% CI (1.18–5.31); p=0.017) is associated with the development of circulatory system diseases in experienced miners with occupational dust related lung pathology. For the wild allele T (OR=0.59; 95% CI (0.35–0.99); p=0.048) and the homozygous T/T genotype (OR=0.33; 95% CI (0.15–0.73); p=0.017) of the T704C polymorphism of the AGT gene, a protective effect against the development of cardiovascular pathology was detected in the studied occupational cohort of individuals.Limitations. The study was limited to a small sample of patients.Conclusion. The results of the study indicate the predisposition of miners with the mutant C allele and the heterozygous T/C genotype of the T704C polymorphism of the AGT gene to the formation of circulatory system diseases against the background of occupational lung pathology. The normal T allele and the homozygous T/T genotype are associated with the resistance to the development of cardiovascular pathology in long-term labour duration miners.Ethics.
