Estimation of FTA-card Suitability of semen samples for DNA profile after long- time Preservation
Background: Cards from Flinders Technology Associates (FTA) simplify sample storage, transport, and extraction while cutting costs and diagnostic time. On rare occasions, the regular operation may fail to provide a complete STR profile. This research aimed to evaluate the FTA's suitability for the secure conveyance and preservation of human seminal fluid material before and after long- time preservation of samples on FTA cards and estimate complete DNA profile generated by using AmpF‘STR® MiniFiler Kit. Methodology: Twenty-five semen samples were collected from volunteers. DNA extraction was carried out by using the organic method. Using the Applied Biosystems AmpFiSTR Identifiler kit, fifteen STR markers were genotyped, the Genetic Analyzer and Gene Mapper ID were used to perform capillary electrophoresis and data analysis. Results: The findings showed that there was a substantial difference in the DNA concentration and purity, respectively. The allele percentage appears complete and partial STR profiles (100%) (66.6%,53.3%, 60%,73%,73%,66.6%,53.3%) respectively in cases before and after preservation. Conclusion: The FTA card approach was determined to be a reliable method for extracting and purifying DNA from human seminal fluid but causes DNA deterioration over time. Therefore, it is advisable to extract DNA from the FTA card as soon as it is practical after collection.
