In Vitro Effects of Mir-6757-3p Modulation and AT9283 Treatment on AURKA/AURKB Expression and Apoptosis-Related Responses in A549 and BEAS-2B Cells

Background: Non-small cell lung cancer (NSCLC) is one of the leading causes of cancer-related deaths. MicroRNAs (miRNAs) are important regulators of tumor biology, and Aurora kinases (AURKA, AURKB) play a critical role in the cell cycle and tumor development. Objective: This study aimed to investigate the association of miR-6757-3p modulation with Aurora kinase expression and apoptosis-related responses under in vitro conditions. Methods: A549 and BEAS-2B cells were treated with miR-6757-3p mimic, inhibitor, and the multi-target kinase inhibitor AT9283. Gene expression levels of BAX, BCL2, CASP3, CASP9, MTOR, AURKA, and AURKB were determined by qRT-PCR. miR-6757-3p expression was analyzed separately using a miRNA-specific qRT-PCR assay. Findings: Changes in the expression of apoptosis-related markers were observed following miR-6757-3p modulation and AT9283 treatment. These findings reflect treatment-associated molecular responses and do not, by themselves, establish the occurrence or extent of apoptosis. Changes in MTOR expression were also observed following treatment. Protein-level analyses showed partially consistent patterns with the corresponding gene expression findings, although not all changes reached statistical significance. Time-dependent wound-healing analyses revealed differences in wound closure following miR-6757-3p inhibition, miR-6757-3p mimic treatment, and AT9283 treatment. Conclusions: miR-6757-3p modulation was associated with changes in AURKA/AURKB expression and apoptosis-related markers in A549 and BEAS-2B cells. These findings provide preliminary in vitro evidence for an association between miR-6757-3p modulation, Aurora kinase expression, and apoptosis-related responses. Further mechanistic and in vivo studies are required.
