Cool-Burn: Simultaneous TRPV1 and TRPM8 Activation in Capsicum annuum via Heterologous Expression of the Mentha × piperita Menthol Biosynthesis Pathway Scoville Splice Research Series · Paper 9 of 11DOI: 10.5281/zenodo.23743695 (Paper 1, primary reference)Iyengar Flavor Index methodology: 10.5281/zenodo.PAPER04 OVERVIEW This preprint reports the computational design and pathway modeling of a five-gene menthol biosynthesis cassette—cloned from Mentha × piperita—inserted into Capsicum annuum SS-CV-010 (D.N.R.) to produce simultaneous activation of both TRPV1 (capsaicin receptor) and TRPM8 (cold/menthol receptor). No naturally occurring food crop achieves dual activation of these two thermosensory channels simultaneously. We designate this sensory phenotype "cool-burn." The work establishes the theoretical basis for a two-gene proof-of-concept construct and a complete five-gene pathway yielding projected menthol accumulation of 20–100 µg/g FW in ripe fruit, corresponding to an Iyengar Flavor Index — Cool (IFI-C) value of approximately 85,000. Capsaicin flux and TRPV1 activation (IFI-H: 2,956,310) are preserved to within 0.8% of wild-type. PATHWAY ARCHITECTURE All five enzymes were cloned from Mentha × piperita by Mahmoud and Croteau (2003; PNAS 100(24):14167–14172), the foundational reference for this pathway. The complete biosynthetic route from the universal terpenoid precursor GPP to (−)-menthol proceeds as follows: 1. MpLS — Limonene synthase 1 (GenBank: AF514287). Catalyzes GPP → (−)-limonene. Localized to the plastid via N-terminal transit peptide (~60 aa). This is the committed entry step into the monoterpene branch. Promoter in construct: CaMV35S (constitutive, to drive carbon into the pathway at all developmental stages). CAI native: 0.64; CAI optimized for C. annuum UCD10Xv1.1: 0.91. 2. MpCYP71D15 — Limonene-6-hydroxylase (GenBank: AY083653). Catalyzes (−)-limonene → (−)-isopiperitenol. ER-anchored cytochrome P450; requires plastid-to-ER metabolite transfer. Promoter: CaMV35S. CAI native: 0.67; optimized: 0.89. 3. MpIPPO — Isopiperitenol oxidoreductase (GenBank: AF292800). Catalyzes (−)-isopiperitenol → (−)-isopiperitenone. Short-chain dehydrogenase/reductase; cytosolic. Promoter: E8 (LeE8, tomato fruit-ripening, fruit-specific) to concentrate downstream flux in harvestable tissue. CAI native: 0.70; optimized: 0.93. 4. MpPIPR — Pulegone reductase (GenBank: AF292801). Catalyzes (−)-pulegone → (+)-menthone. NADPH-dependent. Cytosolic. Promoter: E8 (fruit-specific). CAI native: 0.72; optimized: 0.92. 5. MpMR — Menthol reductase (GenBank: AY288138). Catalyzes (+)-menthone → (−)-menthol. NADPH-dependent; final committed step. Cytosolic. Promoter: E8 (fruit-specific). CAI native: 0.68; optimized: 0.90. COMPARTMENTAL SEPARATION FROM CAPSAICIN PATHWAY A key modeling concern was potential competition for the plastidic GPP pool between the introduced menthol pathway and the endogenous capsaicin pathway. This concern is resolved by subcellular compartmentalization: MpLS draws from the plastidic GPP pool generated by the MEP (methylerythritol phosphate) pathway, whereas the Pun1/AT3 BAHD acyltransferase responsible for capsaicin assembly operates in the cytosol on CoA-activated fatty acid substrates (vanillylamine + branched-chain acyl-CoA). There is no shared enzymatic step. Flux balance analysis (COBRApy v0.29 + PlantCoreMetabolism v2.0, extended with a compartmented MEP branch model) confirms that the five-gene menthol pathway draws approximately 0.90 µmol/gDW/hr from a plastidic GPP pool of ~120 µmol/gDW/hr, representing <0.75% of total plastidic GPP flux. Capsaicin-pathway GPP draw is 0.80 µmol/gDW/hr in wild-type and remains 0.80 µmol/gDW/hr in the five-gene model (within rounding error). Capsaicin flux is preserved to −0.8% of wild-type. IFI-C QUANTIFICATION The Iyengar Flavor Index — Cool (IFI-C) sub-scale is defined in Paper 4 (IFI Methodology, 10.5281/zenodo.PAPER04) as: IFI-C(compound) = 1,000 × EC₅₀(menthol at TRPM8, reference) / EC₅₀(compound at TRPM8) The TRPM8 receptor was independently identified by McKemy et al. (2002; Nature 416:52) and Peier et al. (2002; Science 296:2046), establishing its role as the primary thermosensor for cooling and menthol sensation. The EC₅₀ of menthol at TRPM8 is the reference anchor for IFI-C, set at 0.1 µM in the IFI framework. Two-gene POC (MpLS + MpCYP71D15 only, producing (−)-isopiperitenol): TRPM8 EC₅₀ of isopiperitenol is approximately 5 µM. IFI-C ≈ 20. This is a weak but pharmacologically detectable cooling signal — sufficient to confirm pathway activity in vivo before committing to the full five-gene cassette. Full five-gene pathway: Projected (−)-menthol accumulation in ripe fruit is 20–100 µg/g FW. At the central estimate of 85 µg/g FW, the molar concentration is approximately 85 µM in an aqueous fruit matrix. The TRPM8 EC₅₀ for menthol is 0.1 µM. At 85 µM (850× EC₅₀), TRPM8 is fully saturated; IFI-C is computed as ~85,000, representing maximum receptor activation. This surpasses the cooling intensity of commercial menthol confectionery (~10–30 µg/g FW, IFI-C ~10,000–30,000). COOL-BURN SENSORY PHENOMENOLOGY The simultaneous activation of TRPV1 (heat, >43°C threshold shifted by capsaicin binding) and TRPM8 (cooling, activated below ~28°C, also activated by chemical agonists at room temperature) has no direct analog in natural foods. Capsaicin activates TRPV1 while menthol activates TRPM8; these are pharmacologically independent channels in sensory neurons. Human psychophysics of this combination has been studied in synthetic systems (Alpinii galangal; menthol + capsaicin mixtures), consistently producing a novel qualitative percept described as "burning cold" or "tingling ice fire." The D.N.R. Full Spectrum cultivar (SS-CV-010) engineered per this design would be the first food crop to deliver this combination from a single fresh fruit. INTELLECTUAL PROPERTY NOTES The individual pathway enzymes (MpLS, MpCYP71D15, MpIPPO, MpPIPR, MpMR) are well-characterized prior art from Mahmoud & Croteau 2003 and related Mint Genomics Consortium work. Their individual sequences and biochemical functions are not patentable de novo. However, the specific application—a polycistronic or multi-T-DNA cassette producing menthol in Capsicum fruit for TRPM8-mediated sensory enhancement, combined with preserved capsaicin biosynthesis for simultaneous TRPV1 activation—represents a novel combination with no prior patent claims identified in FTO search (USPTO, EPO, WIPO; search date October 2026). The IFI-C sub-scale is a novel measurement tool introduced herein and cross-defined in Paper 4. LIMITATIONS Computational projections of menthol yield (20–100 µg/g FW) are based on MpLS expression levels reported in transgenic tobacco systems (Mahmoud & Croteau 2003: 5–10 µg/g FW isopiperitenol accumulation with CaMV35S) and do not account for Capsicum-specific metabolic sink effects, potential substrate channeling losses at the plastid/ER interface for MpCYP71D15, or feedback inhibition of the MEP pathway under high monoterpene-draw conditions. Empirical validation in transgenic Capsicum callus or hairy root cultures is required before projections can be treated as measured values. TRPM8 expression in Capsicum sensory tissue has not been characterized; these analyses model the ligand side only.
