Impact of in vivo static magnetic field exposure on a single nucleotide polymorphism in exons 7 and 8 of the rat Trp53 gene

Aim: Static magnetic field (SMF) exposure and its influence on genetic mutations is a global concern. This study aimed to investigate the effects of SMF exposure on the Trp53 gene mutations of albino rats. Methods: Three groups of albino rats participated. Groups A and B were exposed in-vivo to a 2.4 mT SMF: Groups A and B were exposed to SMF 1 h/day for 3 weeks, and 4 h/day for 2 weeks, respectively. A control group (Group C) was left unexposed. Blood samples were collected after exposure and genomic DNA was extracted from the whole blood samples. Two primers targeting exons 7 and 8 of the Trp53 gene were designed and used for PCR amplification. The PCR products visualized using agarose gel electrophoresis technique, then sequenced. The process of sequence analysis was accomplished with BioEdit, FinchTV, and Mutation Surveyor. Reference sequences were obtained from the NCBI database. Results: Group A (1 h/day exposure) revealed Mutations, both of deletion and insertion were induced in exon 7 of the Trp53 gene. Consequently, the nucleotide sequence has altered between positions 32–45, these alterations resulting in amino acid variations between residues 198–211. No mutation was identified from Group B samples (4 h/day). Conclusion: Mutations in the Trp53 gene of rats were produced after repeated short-term daily SMF exposure (1 h/day) for three weeks. “These results suggest that SMFs can induce genomic instability in tumor suppressor genes, although further large-scale and long-term studies are needed to confirm this finding.
