Pengaruh Nanokurkumin Terhadap Siklus Estrus Tikus Model Pcos
ABSTRACT Polycystic ovarian syndrome (PCOS) is a hormonal illness that affects women of reproductive age. It is distinguished by elevated testosterone levels, insulin resistance, and menstrual cycle irregularities. Kurkumin delivered by a Self-Adhesive Nanoemulsification Drug Delivery System (SNEDDS) has been proposed as an additional therapy to increase its availability and therapeutic efficacy. The purpose of this experimental investigation was to determine the effect of SNEDDS kurkumin on improving the estrous cycle in a well-established rat model of PCOS. The methodology employed was a control group design with post-experiment testing performed in vivo on female Rattus norvegicus Wistar strain rats that had been induced with PCOS using letrozole and a high-fat, high-fructose diet for 21 days. The experimental animals were placed into six groups: normal control, negative control, positive control (metformin), and three groups that received SNEDDS kurkumin at varied oral doses for 14 days. Daily vaginal smear cytology was used to investigate estrous cycles, with One-Way ANOVA and Tukey's post hoc test (α = 0.05) applied. The PCOS group experienced significantly longer estrous cycles than the normal control group and all treatment groups. Administration of SNEDDS kurkumin reduced the duration of estrous cycles in a dose-dependent manner compared to the negative control and metformin groups, indicating improved regulation of the reproductive cycle in the treated animals. SNEDDS kurkumin effectively shortened estrous cycle duration in the PCOS rat model, suggesting its potential as an adjuvant therapy for reproductive cycle disturbances associated with PCOS.
