Cracking the miRNome: lessons learned and future directions in microRNA CRISPR screening
Abstract While microRNA are central regulators of cellular biology, their therapeutic translation has lagged behind that of protein-coding genes, in part due to a lack of comprehensive functional interrogation. Performing microRNA-focused CRISPR-Cas9 screens is crucial to define their context-dependent roles in disease. We highlight the methodological overlaps and unique challenges between microRNA and protein-coding gene screens. This review serves as a guiding framework for the design and implementation of microRNA-focused screens.
