BCAT1 in colorectal cancer dissemination: Compartmentalized metabolism and the UMP-vimentin interaction

Branched-chain amino acid transaminase 1 (BCAT1) connects nitrogen metabolism with cytoskeletal remodeling in colorectal cancer models.Cytosolic BCAT1 transfers branched-chain amino acid-derived nitrogen through glutamate and aspartate to uridine monophosphate (UMP).UMP binding reduces vimentin ubiquitination and proteasomal turnover, supporting migration and dissemination.Mitochondrial BCAT activity instead favors glutamate dehydrogenase-dependent ammonia formation in the studied cells.A single primary study reports this sequence in colorectal cancer cell lines and mice, alongside retrospective human expression and survival associations. This review examines compartmentalized nitrogen metabolism, vimentin regulation, epithelial and metastatic niches, circulating methylated DNA biomarkers, and therapeutic implications.Nutrient supply, microbial metabolism and disease stage contribute to contrasting BCAT isoform phenotypes.Blood BCAT1/IKZF1 methylation assays measure tumor-associated DNA release and have been investigated for detection and surveillance.Translation of the metabolic mechanism requires patient-derived replication, quantitative intracellular UMP occupancy, binding stoichiometry and selective pharmacological intervention.Clinical utility for BCAT1-directed treatment remains unproven.Relating endogenous enzyme localization to nitrogen transfer, vimentin engagement and invasion in patient-derived tumors could identify metabolic dependencies for therapeutic investigation, with host muscle, liver and immune function informing treatment safety.

Abstract

Branched-chain amino acid transaminase 1 (BCAT1) connects nitrogen metabolism with cytoskeletal remodeling in colorectal cancer models.Cytosolic BCAT1 transfers branched-chain amino acid-derived nitrogen through glutamate and aspartate to uridine monophosphate (UMP).UMP binding reduces vimentin ubiquitination and proteasomal turnover, supporting migration and dissemination.Mitochondrial BCAT activity instead favors glutamate dehydrogenase-dependent ammonia formation in the studied cells.A single primary study reports this sequence in colorectal cancer cell lines and mice, alongside retrospective human expression and survival associations. This review examines compartmentalized nitrogen metabolism, vimentin regulation, epithelial and metastatic niches, circulating methylated DNA biomarkers, and therapeutic implications.Nutrient supply, microbial metabolism and disease stage contribute to contrasting BCAT isoform phenotypes.Blood BCAT1/IKZF1 methylation assays measure tumor-associated DNA release and have been investigated for detection and surveillance.Translation of the metabolic mechanism requires patient-derived replication, quantitative intracellular UMP occupancy, binding stoichiometry and selective pharmacological intervention.Clinical utility for BCAT1-directed treatment remains unproven.Relating endogenous enzyme localization to nitrogen transfer, vimentin engagement and invasion in patient-derived tumors could identify metabolic dependencies for therapeutic investigation, with host muscle, liver and immune function informing treatment safety.
