Evaluation of The Effectiveness of Melastoma malabathricum L. Ethanol Leaf Extract Cream on Incision Wound Healing in Male White Mice (Mus musculus)

Background: Incision wounds require appropriate treatment to reduce infection risk and promote tissue repair. Melastoma malabathricum leaves contain flavonoids, tannins, saponins, alkaloids, and terpenoids that may contribute to anti-inflammatory, antioxidant, and antimicrobial effects. Objective: This study investigated the wound-healing activity of ethanolic extract cream from M. malabathricum leaves at different concentrations in mice. Methods: Twenty-five male mice were assigned to five groups: Bionect® as the positive control, cream base as the negative control, and 2.5%, 5%, and 7.5% extract cream groups. A 1.5-cm-long and 2-mm-deep incision was made on the dorsal skin and treated once daily for 14 days. Wound length was assessed on days 3, 7, 10, and 14, followed by One-Way ANOVA and LSD analysis (α = 0.05). Results: The extract contained flavonoids, alkaloids, tannins, saponins, and terpenoids. All cream preparations met the physical quality requirements and produced no skin irritation. On day 14, wound lengths were 0.43 ± 0.03 cm, 0.37 ± 0.02 cm, and 0.25 ± 0.02 cm for the 2.5%, 5%, and 7.5% groups, respectively. Healing percentages were 71.00%, 75.17%, and 83.00%, respectively, while the positive and negative controls showed 100% and 66.33%. Significant differences were observed among groups (p < 0.05). Conclusion: Ethanolic extract cream of M. malabathricum leaves promoted incision wound healing in mice, with the 7.5% concentration producing the greatest effect among the tested formulations.
