Neisseria gonorrhoeae : a historical model for host adaptation

ABSTRACT Research into Neisseria gonorrhoeae (Gc) physiology, genetics, and pathogenicity remains deeply shaped by its history. Long before molecular tools were available, early clinical observations, human inoculation studies, and initial vaccine trials defined the field’s primary experimental questions and limitations. Key features of gonococcal infection, including restriction to the human host, rapid phenotypic alteration during culture, and the absence of durable protective immunity, were documented empirically decades before researchers understood their underlying biological mechanisms. As a result, contemporary gonococcal research has largely developed in response to these early empirical findings. Modern genomic and molecular studies have provided mechanistic explanations for long-standing clinical observations, turning historical paradoxes into defined areas of investigation. From designing vaccines that target variable surface structures to surveilling horizontally acquired resistance determinants, the questions driving current research directly continue the experimental challenges investigators identified.
