VISUAL INSPECTION TO QUANTITATIVE IMAGE ANALYSIS: COMPUTER VISION IN MICROBIOLOGY

Microbiology has historically relied on visual interpretation of microscopic images to infer microbial structure, function, and classification. While foundational, many classical interpretations were shaped by limitations in imaging technology, sample preparation artefacts, and subjective human judgement. As a result, visually compelling but biased observations were sometimes accepted as biological reality. This review revisits classical microbiological interpretations through the lens of computer vision and image analysis. Rather than focusing on algorithmic novelty, the paper reframes long-standing microbiological challenges as fundamental computational problems, including segmentation, classification, measurement, and noise and bias handling. Representative examples—such as mesosomes, cell wall thickness variability, spore layer ambiguity, appendage misclassification, biofilm architecture, inclusion bodies, viability assessment, morphology-based taxonomy, and colony counting—are examined to illustrate how image-driven misinterpretations arose and how quantitative analysis resolves them. By synthesizing literature from microbiology and computer science, this review highlights how treating microbial images as data rather than illustrations enables objective, reproducible, and scalable interpretation. The paper emphasizes that computer vision does not replace biological insight but strengthens it by enforcing consistency and transparency. This interdisciplinary perspective supports more reliable microscopy-based conclusions and helps prevent the recurrence of historical misinterpretations in microbial research.
