TileBac - an annotated ultralow-dose cryo-EM montage of bacterial membranes and flagella

Abstract Large-area cryogenic electron microscopy (cryo-EM) requires a tradeoff among field of view, acquisitiontime, and electron exposure. The goal is to mitigate this tradeoff by quickly collecting large fields of viewwith enough dose to detect features of interest using segmentation models but low enough dose to limitsample damage. The dataset presented here, TileBac, comprises two annotated cryo-EM datasets of Pantoeasp. YR343 collected with imaging conditions to achieve this goal. The first contains 2048 × 2048 pixel tilesderived from wide-field montages acquired at a total dose of 4 e−/Å2 with manual pixel-level annotationsof inner- and outer-membrane-enclosed regions. The second contains images acquired at a total dose of40 e−/Å2, with bacteria and flagella annotated as separate classes. These corresponding 40-frame moviesretain individual 1 e−/Å2 frames, allowing users to construct dose-dependent image series from the sameexposure. Images and annotations are distributed in standard MRC, PNG, YOLO, and COCO formats. Therecords are intended to support development and evaluation of segmentation, registration, stitched inference, and dose-dependent image-analysis methods for low-signal cryo-EM.
