Fluorinated Fatty Acids: Origin, Synthesis and Activity

Fluorinated fatty acids occupy a distinctive position at the intersection of natural-product chemistry, lipid metabolism, and synthetic organofluorine chemistry. This mini-review examines their natural occurrence, structural characterization, proposed biosynthetic origins, selected synthetic approaches, and biological effects, with particular emphasis on plant-derived ω-fluorinated fatty acids. Fluorinated constituents of Dichapetalum toxicarium seed oil illustrate variation in chain length and unsaturation, while historical assignments highlight the importance of analytical confirmation. A biosynthetic model involving fluoroacetyl-CoA as a starter unit offers an explanation for terminal fluorination, although precursor transport, enzyme specificity, and individual transformations require distinction between direct evidence and inference. Biological consequences are considered in relation to compound structure and metabolic fate, including experimental evidence for conversion of a terminally fluorinated fatty acid to fluorocitrate. Selected synthetic examples encompass terminal radiofluorination, fluorinated coriolic-acid analogues, and 22-[18F]fluorodocosahexaenoic acid, a tracer evaluated preclinically for brain fatty-acid uptake. Nonradioactive analogues and radioactive tracers are discussed separately, and activities of parent lipids are distinguished from those demonstrated for fluorinated derivatives. The available evidence supports compound-specific interpretation rather than universal claims concerning toxicity, metabolic stability, or therapeutic potential. Priorities include verification of historical structures, characterization of plant biosynthetic pathways, quantitative comparative bioassays, and identification of metabolic products.
