Newcastle disease virus (NDV), a member of the Paramyxoviridae family, is a highly contagious avian pathogen that causes substantial economic losses in the poultry industry. The hemagglutinin-neuraminidase (HN) gene plays a critical role in viral attachment to host cells, viral entry, and the induction of immunogenic properties, making it an important target for molecular studies. In this study, the HN gene of NDV was amplified from viral RNA extracted from the allantoic fluid of infected chicken embryos. Complementary DNA (cDNA) was synthesized by reverse transcription, and the full-length HN gene fragment (~1.7 kb) was subsequently amplified by polymerase chain reaction (PCR). The purified PCR product was cloned into a plasmid vector using a commercial cloning system and transformed into competent Escherichia coli cells. Recombinant colonies were verified by colony PCR and restriction enzyme analysis. The results confirmed the successful amplification and molecular cloning of the HN gene. The obtained recombinant constructs provide a valuable tool for investigating the genetic structure, antigenic characteristics, and virulence determinants of NDV at the molecular level. Furthermore, these findings may contribute to the development of novel diagnostic approaches and vaccine strategies, as well as support future studies on the pathogenicity of NDV infection.
