Keyora Skin Systems Nutrition - Collagen Tripeptides EP-8: The Skin Elasticity and Wrinkle Evidence Matrix: Peptide Identity, Biomechanical Measurement, and Randomized Human Outcomes

Background Skin firmness, elastic recoil, viscoelastic recovery, and periocular wrinkle morphology are related but noninterchangeable outcomes of skin aging. Fibrillar collagen contributes tensile resistance, elastic fibers support recoil, and facial expression, anatomical site, ultraviolet exposure, and baseline phenotype modify observable changes. Clinical evaluation of oral collagen peptides therefore requires distinct mechanical and morphological endpoints. Objective and Scientific Framework This narrative evidence synthesis develops the Keyora Skin Elasticity-and-Wrinkle Evidence Matrix, linking peptide preparation identity, experimental biology, randomized human comparisons, quantitative outcomes, observation time, population context, and methodological quality. It distinguishes Cutometer gross (R2 = Ua/Uf), net (R5 = Ur/Ue), and biological (R7 = Ur/Uf) elasticity from wrinkle volume, surface roughness, and visual severity. Mechanistic Foundations Human studies detected circulating hydroxyproline-containing peptides, including Pro-Hyp, after ingestion of gelatin hydrolysates at 9.4-23 g, with peptide-form hydroxyproline peaking approximately 1-2 hours postdose. In cultured human dermal fibroblasts, Pro-Hyp at 200 nmol/mL increased proliferation approximately 1.5-fold, hyaluronic acid synthesis 3.8-fold, and HAS2 mRNA expression 2.3-fold; human skin explant experiments separately examined matrix-related responses. These models support biological plausibility, not confirmed skin concentrations or efficacy. Experimental and Human Evidence Two separate placebo-controlled Proksch trials reported significant elasticity improvements in a 69-woman trial comparing 2.5 or 5 g/day with placebo for eight weeks and approximately 20% lower periocular wrinkle volume versus placebo in a separate 114-woman trial comparing 2.5 g/day with placebo at week eight. In a wrinkle-trial subgroup, type I procollagen was approximately 65% higher and elastin 18% higher, whereas the 6% fibrillin difference was nonsignificant. Kim (2018) investigated 1 g/day tripeptide-enriched fish collagen for twelve weeks: Cutometer R2 (0.76 versus 0.73; p = 0.025) and R5 (0.71 versus 0.64; p = 0.027) differed significantly from placebo, but R7 did not (p = 0.139). CPNS (1.65 g/day) produced significant selected elasticity findings at week twelve and instrumental wrinkle differences at weeks four and twelve, but not eight. Tripep20 (1 g/day) showed significant R2/R5/R7 and Ra/Rq differences by week six; visual crow's-feet scores and Rmax became significant at week twelve. Integrated Scientific Interpretation Women's concerns about firmness, resilience, or crow's-feet are matched to independently measured outcomes, not presumed interchangeable benefits. Keyora specifies 1,500 mg tilapia-scale collagen peptide powder per two tablets, a declared 30% total tripeptide fraction, and approximately 459 Da supplier-reported mean molecular weight. Other ingredients include elastin peptide, hyaluronic acid, ceramide NP, vitamin C, nicotinamide, and biotin. The 30% fraction represents total tripeptides, not a measured Gly-Pro-Hyp sequence proportion. These specifications establish research identity, not clinical equivalence or demonstrated formulation synergy. Evidence Boundaries and Conclusions A 2025 meta-analysis of 23 trials (1,474 participants) found overall benefits but no significant improvement in higher-quality or non-pharmaceutical-funded subgroups; a 2026 review of 25 trials also raised risk-of-bias concerns. The matrix preserves preparation, comparator, dose, endpoint, and time specificity. Mechanistic observations, external trials, and supporting ingredients cannot validate the complete Keyora formulation or establish universal effectiveness.

Fibrillar collagen contributes tensile resistance, elastic fibers support recoil, and facial expression, anatomical site, ultraviolet exposure, and baseline phenotype modify observable changes.
