Genome-wide analyses are currently widely used in animal husbandry to identify genetic markers of economically valuable traits. DNA extraction is the initial step in genome-wide studies, significantly affecting the genotyping results and the reliability of the obtained results. Therefore, the quality of the isolated DNA intended for further genome-wide studies is of particular importance. This study provides a comparative analysis of DNA extraction methods from frozen sheep blood using two types of commercial kits and an assessment of the suitability of the isolated DNA for genome-wide studies. Additional steps in the preliminary preparation of sheep blood samples are described, significantly improving the quality and purity of DNA isolated from this type of biological material. Results showed that the concentration and spectral characteristics of DNA isolated using the modifications proposed by us were superior to those obtained using standard methods. DNA extracted using the modified method proposed by us is suitable for further microarray genotyping and allows for obtaining high-quality results. The methodological approaches presented in the work do not require additional financial costs and can be used for the successful extraction of DNA from problematic samples of various types of biological material.
