Porcine reproductive and respiratory syndrome virus (PRRSV) remains one of the most economically important pathogens of industrial pig production. Two PRRSV strains representing the major genotypes, Arterivirus/LKZ/2010 (PRRSV-2) and Kostanay-CM/08 (PRRSV-1), were propagated in MARC-145 cells. Clarified virus-containing suspensions were treated with binary ethylenimine (BEI/DEI) at final concentrations of 0.01%, 0.05%, and 0.10% at 22 ± 2°C. Infectious activity was determined at baseline and then at 3-h intervals during treatment; experiments were performed in triplicate (n = 3). Residual inactivant was neutralized with sodium thiosulfate, and completeness of inactivation was confirmed by three consecutive blind passages in MARC-145 cells. Results. For Arterivirus/LKZ/2010, complete loss of detectable infectivity was observed at 30, 24, and 18 h at 0.01%, 0.05%, and 0.10% BEI/DEI, respectively. For Kostanay-CM/08, the corresponding endpoints were 27, 21, and 15 h. Kinetic curves demonstrated a concentration-dependent decline in infectious titer for both strains. No cytopathic effect was detected during three blind passages of samples corresponding to complete inactivation. Increasing BEI/DEI concentration accelerated PRRSV inactivation under the tested conditions. This study assessed loss of infectivity only; antigenicity and immunogenicity were not experimentally evaluated and therefore require separate investigation before selection of a final vaccine-production regime.
