The Diagnostic Value and Molecular Mechanism of miR-1271-5p Combined with Ultrasound in Endometriosis

Endometriosis (EMs) is a systemic inflammatory disease characterized by the growth of endometrial tissue outside the uterus. Its underlying mechanisms remain unclear, contributing to limitations in diagnosis and treatment. This study evaluated the diagnostic value of serum miR-1271-5p combined with transvaginal ultrasound (TVUS) and examined the regulatory relationship between miR-1271-5p and insulin-like growth factor 1 receptor (IGF1R). The study included 115 patients with EMs and 90 healthy volunteers. MiR-1271-5p and IGF1R expression levels were measured using real-time quantitative polymerase chain reaction (RT-qPCR). Receiver operating characteristic (ROC) analysis was used to assess the diagnostic performance of miR-1271-5p and TVUS individually and in combination. A dual-luciferase reporter assay was used to verify the interaction between miR-1271-5p and IGF1R, and in vitro experiments were conducted using hEM15A and 12Z cells. The combined use of miR-1271-5p and TVUS demonstrated high diagnostic performance for EMs. MiR-1271-5p negatively regulated IGF1R, and inhibition of miR-1271-5p significantly reduced cell proliferation, migration, and invasion. Silencing IGF1R reversed these effects. Serum IGF1R mRNA expression was reduced in patients with EMs and was inversely correlated with miR-1271-5p expression. These findings support the diagnostic value of serum miR-1271-5p combined with TVUS and indicate that miR-1271-5p regulates EMs-associated cellular behavior through IGF1R.
