Programmable illumination and spatially varying aberration correction in a laser scanning confocal microscopy system

Abstract Laser scanning confocal microscopy (LSCM) enables high-resolution optical sectioning and three-dimensional imaging through point illumination and confocal detection. However, conventional galvo scanner mirror-based LSCM is constrained by limited beam repeatability, complex alignment, and optical aberrations, while programmable LSCM systems employing spatial light modulators or digital micromirror devices suffer from beam-quality degradation due to grating aperiodicity, non-uniform diffraction efficiency, and beam tilt. Furthermore, both configurations are affected by spatially varying (anisoplanatic) aberrations that cannot be effectively corrected using conventional adaptive optics based on the isoplanatic assumption. In this work, we propose high-quality programmable beam-generation techniques that produce uniform intensity, accurately positioned illumination beams while mitigating grating-induced aperiodicity. We further introduce an astigmatic spot-based wavefront sensing method for sensitive, localized estimation of anisoplanatic aberrations, enabling adaptive correction across the entire field of view. Simulation and experimental results demonstrate that the astigmatic spot provides up to $\sim 17 \times$ higher sensitivity than the conventional DL spot in the low-aberration regime. The proposed multiplexed grating-based scanning enables a nine-fold increase in scanning speed and the anisoplanatic aberration correction improves the resolution by $\sim 20.6\%$ at a contrast threshold of 0.25. Simulation and experimental results demonstrate improved beam quality, aberration correction, image contrast, and overall imaging performance, establishing a robust framework for next-generation programmable confocal microscopy.

In this work, we propose high-quality programmable beam-generation techniques that produce uniform intensity, accurately positioned illumination beams while mitigating grating-induced aperiodicity.
