Membrane state may tune calcium's restraint of renin secretion

This Mini-Review examines the hypothesis that membrane organisation in a defined juxtaglomerular compartment conditions the strength and timing of calcium-mediated restraint of renin secretion. Within the membrane-encoded chemosensory system (MECS) framework, cholesterol accessibility, lipid organisation or membrane mechanics could influence calcium recruitment, calcium-dependent cyclic AMP suppression or downstream secretion. If inhibitory coupling is affected, comparable relevant local calcium dynamics could produce different active-renin responses. The manuscript develops complementary predictions for pressure dependence and active-renin recovery, which may change independently. Similar initial suppression during a pressure pulse could precede different recovery after pressure returns to its starting value, potentially altering cumulative active-renin output. A separate reversibility prediction is that changing a measured membrane property should alter the affected signalling contribution and renin response, while restoring that property before a matched challenge should recover both. Cannabinoid, olfactory and TRPV4-associated routes are examined as candidate implementations, including a hypothetical arrangement involving vascular mechanics and a separate mouse Olfr78 metabolite input. The review distinguishes established experimental precedents from proposed mechanisms. Native membrane mediation and combined route operation remain untested in the juxtaglomerular apparatus. Potential consequences for renin–angiotensin–aldosterone signalling and clinical responses require independent validation, including human evidence.
