Mitochondrial Electron Flow Modulation and the "Cellular Panic" Hypothesis: Therapeutic Implications in Cancer, Neurodegeneration, and Aging
Abstract: Classical oncology largely attributes carcinogenesis to accumulated somatic nuclear mutations. However, growing bioenergetic evidence indicates that primary mitochondrial metabolic dysfunction serves as an initiating upstream event. Herein, we present the "Cellular Panic" hypothesis, linking oxidative stress, mitochondrial respiratory chain failure, and cytosolic mtRNA/mtDNA leakage to chronic sterile inflammation and epigenetic instability. To intervene at this metabolic juncture without the pitfalls of non-specific antioxidants, we designed the Mitochondrial Quantum Tuner (MQT) series as precision Electron Flow Modulators. Through molecular docking against human respiratory Complex I (PDB: 9i4i) and Complex III (PDB: 9hzl), the MQT lead prototype demonstrated robust binding affinities (-6.546 kcal/mol and -7.553 kcal/mol) with highly stable interaction profiles. Furthermore, we outline context-dependent therapeutic applications ranging from selective cancer cell elimination to neuroprotection in Parkinsonian models.
