PFEP-Assisted Intramolecular Disulfide Cyclization of Setmelanotide
Setmelanotide is the octapeptide Ac-Arg-Cys-D-Ala-His-D-Phe-Arg-Trp-Cys-NH2 cyclized by a single Cys2–Cys8 disulfide. The compact sequence contains two D-residues, multiple basic residues, and oxidation-sensitive Trp. PFEP is proposed as a low-dose oxidation and immediate-quench platform that maximizes intramolecular closure while limiting dimerization and aromatic oxidation. A concentration-dependent model predicts 88.2% cyclic monomer at 0.30 mg/mL under programmed dosing versus 73.6% for a bolus comparator; synthetic Trp-oxidized species are 1.8% versus 5.6%. The data are simulations. The design includes concentration series, matched microdosing, peroxide/metal controls, reduction challenge, LC-HRMS, and MC4R potency. Article type: Target-specific process hypothesis with synthetic cyclization data. All numerical results labelled synthetic or modelled are hypothesis-illustration and planning outputs, not experimental measurements. No wet-laboratory experiment, validated process, clinical benefit, or shelf life is claimed.
