Targeting (K)RAS in pancreatic ductal adenocarcinoma—ASCO 2026 highlights
Summary KRAS mutations are present in approximately 90% of pancreatic ductal adenocarcinomas and have historically been considered undruggable. Daraxonrasib is a first-in-class RAS(ON) inhibitor that recently demonstrated improved survival compared with chemotherapy in the phase 3 RASolute 302 trial. In addition, early-phase data on selective KRAS G12D inhibitors show clinical efficacy with a tolerable safety profile. Ongoing studies are investigating combination strategies and the potential use of RAS inhibitors in first-line and potentially curative settings.
