Traditional glycosylations rely on protecting‐group manipulations, resulting in cumbersome synthetic routes. Starting form unprotected sugars, a new N‐bromosuccinimide (NBS)‐mediated Mitsunobu glycosylation method enabling the direct construction of N‐glycosides was developed. The method was successfully applied to the monosaccharide composition analysis of polysaccharides from traditional Chinese medicine. Abstract
