Benzimidazole Derivatives: Chemistry, Biological Activity, and Therapeutic Applications
Abstract Benzimidazole derivatives are important nitrogen-containing heterocyclic compounds with broad medicinal significance. Their fused benzene-imidazole nucleus provides a versatile platform for structural modification and interaction with diverse biological targets. This review summarizes the major pharmacological actions of benzimidazole derivatives, with emphasis on antimicrobial, antifungal, anthelmintic, antiprotozoal, and anticancer activities. The pharmacological classification, mechanisms of action, drug-resistance aspects, and therapeutic significance of these activities are discussed. Benzimidazole derivatives can interfere with microbial enzymes, fungal sterol pathways, parasite microtubules, protozoal metabolism, and cancer-associated molecular pathways. Several established benzimidazole drugs demonstrate clinically relevant activity, while newer derivatives are being investigated for improved potency and selectivity. Structure–activity relationships, comparative pharmacological features, and future opportunities in medicinal chemistry are also highlighted. Overall, the benzimidazole scaffold remains a valuable starting point for the development of multifunctional therapeutic agents and continues to support research into safer, more selective, and resistance-conscious drug candidates through rational structure-based drug design approaches.
