The Effect of Choline on Viability in Prostate Carcinoma Cells: Possible Involvement of Cholinergic Regulation
Background and Purpose: Prostate carcinoma is a frequently encountered tumor, and due to its relatively long survival period, early diagnosis, treatment, and follow-up are crucial in this cancer. Changes in the tumor microenvironment, including neural alterations, have drawn attention to their effects on treatment and prognosis. The mechanism of neural effects, such as acetylcholine (ACh), in prostate cancer is not yet fully elucidated. The aim of this research is to investigate the possible interaction of choline on cell viability in prostate carcinoma cells. The aim of this study to investigate the effects of choline in normal prostate cells and cells of prostate carcinoma. Methods: DU145 human prostate epithelial carcinoma cells and PNT1a human prostate epithelial normal cell lines were treated with increasing concentrations of choline (0.5-2.5-5-25-50-75-100 µM) and viability analysis using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method.Results: Our preliminary results showed that choline suppressed the viability of prostate carcinoma cells at concentrations of 50µM and higher however, viability in normal prostate cells remained unchanged. Conclusion: Although further analyses are needed, our results demonstrate that choline might have potential therapeutic effects in prostate carcinoma without effecting normal prostate cells.
