INTEGRATED CHEMOPROFILING, HPLC FINGERPRINTING AND α-AMYLASE INHIBITORY PROFILING OF MADHUMEHARI CHURNA: A MULTIDIMENSIONAL APPROACH TO PHYTOCHEMICAL STANDARDIZATION
Madhumehari Churna is a well known traditional, ethnomedicinal, folkiatric polyherbal formulation traditionally used for the management of diabetes and related metabolic disturbances. The present study was designed to establish an integrated phytochemical and analytical profile of Madhumehari Churna using conventional phytochemical screening together with spectroscopic and chromatographic techniques. The formulation contains medicinal plants traditionally associated with antidiabetic activity, including Gymnema sylvestre, Syzygium cumini, Momordica charantia, Trigonella foenum-graecum, Emblica officinalis, Curcuma longa, Azadirachta indica and Tinospora cordifolia. Preliminary phytochemical investigation was carried out to identify major classes of secondary metabolites. Spectroscopic characterization was used to obtain information regarding functional groups and characteristic chemical constituents, while chromatographic analysis was employed for analytical profiling and marker-compound assessment. High-performance liquid chromatography (HPLC) was performed using a Shim- pack C18 column with UV detection at 275 nm. Catechol, tannic acid and quercetin were considered as reference standards for chromatographic characterization and assessment of potential antidiabetic activity by gold standard in-vitro model like porcine pancreatic alpha amylase enzyme inhibition assay. The combined integrated multi-analytical charectrization and standardization approach provides a scientific basis for the identification, standardization and quality assessment of Madhumehari Churna and may support further pharmacological and clinical evaluation of the formulation.
