Deciphering the AGEs-RAGE Signaling Network in Diabetes-Associated Periodontitis: Pathogenic Mechanisms and Emerging Targeted Therapies

Objectives: To summarize the role of the AGEs-RAGE axis in diabetes-associated periodontitis and evaluate potential targeted therapies. Materials and Methods: Recent literature on molecular mechanisms and therapeutic strategies involving the AGEs-RAGE pathway in diabetic periodontitis was reviewed. Results: The AGEs-RAGE axis promotes periodontal inflammation, oxidative stress, and tissue destruction through activation of NF-κB, MAPKs, and NLRP3 inflammasome signaling. Several therapeutic approaches, including RAGE inhibitors, natural compounds, and AMPK activators, have shown potential in preclinical studies. Conclusions: Targeting the AGEs-RAGE pathway represents a potential strategy for managing diabetes-associated periodontitis, although further clinical validation is required.
