A scalable recombinant pipeline for disulphide-stapled peptides — code and data

Analysis code and sequence-free source data for a 96-well recombinant pipeline for disulphide-stapled cyclic peptides: automated IVA primer design, qPCR cloning QC, periplasmic secretion and DOC lysis, IMAC, SEC yield analysis with hierarchical clustering, and Ellman's free-thiol staple QC. Includes the designed expression construct (His6-SUMO-linker-ccdB in pET-24b(+)), the analysis notebooks and figure scripts, source data for Figures 2-5 (including the anonymised per-well SEC traces behind Figure 4), and SPR sensorgrams for the representative designs in Figure 6. Figures 4 and 6 regenerate from the deposited anonymised data with no external inputs; notebook 3 documents the full SEC analysis with the third-party SAPP_DMX engine. See the README for per-figure reproduction notes. Designed peptide sequences are withheld (intellectual property) and binding-target names are anonymised. LICENSING: all code (.py/.ipynb, wherever located) is MIT; all other material (data, construct, protocols, examples, documentation) is CC BY 4.0.
