De novo assembly and characterization of the complete chloroplast genome of Amorphophallus myosuroides Hett. & A. Galloway 2006
The genus Amorphophallus exhibits high species diversity but poorly resolved phylogeny due to phenotypic plasticity and convergence. Amorphophallus myosuroides, with unique silvery leaf markings, lacked complete plastome data, leaving its phylogenetic placement uncertain. We sequenced and assembled its first chloroplast genome using PacBio HiFi sequencing. The circular genome is 171,894 bp in length contains 128 genes, including the trans-splicing gene rps12. Phylogenetic analysis of protein-coding genes from 20 congeneric species robustly placed it in a clade with A. titanum and A. paeoniifolius within the Southeast Asian lineage. This clarifies its evolutionary relationships and provides data for studies on speciation, morphology, taxonomy, and biogeography in Amorphophallus.
