Ferritin Complex Vaccine against Influenza A Using Screened Immunogenic Sequences from Fv-Antibody Library
Abstract A ferritin-based influenza A vaccine was developed. The immunogenic sequences of the nucleoprotein (NP) were screened using an autodisplayed Fv-antibody library, and the paratopes with a high-affinity binding affinity were analyzed through docking simulations. The resulting antigenic sequences were engineered into single-domain constructs using a nonimmunogenic linker. These immunogenic sequences were then expressed in conjunction with ferritin heavy-chain proteins to form spherical nanoparticles through self-assembly. These NP-ferritin complex vaccines were subsequently injected into mice, and antibody responses, cytokine secretion, and T-cell activation profiles were subsequently analyzed to demonstrate that the ferritin-based NP vaccines activated innate as well as acquired immune responses.
