Prognostic value of circ_0049271 in NSCLC and its regulatory effect on tumor
To explore the prognostic significance and functional role of circ_0049271 in non-small cell lung cancer (NSCLC). Clinical data were meticulously collected from 140 patients diagnosed with NSCLC. Lung cancer tissues, along with adjacent normal tissues, were preserved at -80 °C. Additionally, a five-year follow-up was conducted on this patients. qRT-PCR was utilized to quantify the expression levels of circ_0049271, miR-146b-3p and CAMK2B. The CCK-8 and Transwell assay were used to measure the proliferation activity of the cells as well as their migration and invasion capabilities. The binding relationship between the two genes in the circ_0049271/miR-146b-3p/CAMK2B axis was verified by using the dual luciferase reporter assay and RNA immunoprecipitation. circ_0049271 was lowly expressed in lung cancer tissues. Patients with a low expression level of circ_0049271 have a lower survival rate. In contrast, miR-146b-3p showed elevated expression in cancerous tissues and established a direct reciprocal relationship with circ_0049271. Increasing the level of circ_0049271 can inhibit the proliferation and migration of lung cancer cells. However, upregulation of miR-146b-3p counteracted this effect. In NSCLC cells, circ_0049271 upregulates CAMK2B by adsorbing miR-146b-3p. The level of circ_0049271 in NSCLC patients is relatively low. The downregulation of this gene is a risk factor for NSCLC. The reduction of circ_0049271 leads to the down regulation of CAMK2B by releasing miR-146b-3p and accelerates the deterioration of NSCLC.
