Effect of fezolinetant on sleep, health-related quality of life and work productivity

OBJECTIVE: This study aimed to evaluate fezolinetant's impact on sleep disturbance and impairment in women with moderate to severe vasomotor symptoms (VMS) due to menopause and explore relationships between VMS improvement and work productivity. METHOD: This post-hoc analysis of SKYLIGHT 1 and 2 pooled data assessed sleep disturbance and impairment via the Patient-Reported Outcomes Measurement Information System Sleep Disturbance - Short Form 8b (PROMIS SD-SF-8b) and PROMIS SD-SF-8a questionnaires. Correlations were evaluated between improvements in VMS (frequency, severity, frequency at night) and sleep disturbance and Work Productivity and Activity Impairment questionnaire specific to Vasomotor Symptoms (WPAI-VMS) score; and sleep disturbance and work productivity. RESULTS: Fezolinetant 30 mg and 45 mg improved sleep disturbance and impairment versus placebo at 12 weeks, regardless of baseline sleep status. At weeks 12 and 52, both doses showed correlations between VMS frequency/severity and WPAI-VMS productivity scores, and between sleep disturbance and work productivity. CONCLUSION: Fezolinetant reduced moderate to severe VMS due to menopause and improved patient-reported sleep outcomes, with benefits maintained over 1 year. Improvements in VMS and sleep were associated with better work productivity.
