Voltammetric Analysis of DNA in Apricot Varieties
In this study, DNA samples isolated from apricot structures were analyzed using voltammetric analysis, a glassy carbon electrode modified with single-walled carbon nanotubes (SWCNT) capable of easy-to-apply, highly reproducible, and wide-linear measurement. First, DNA was isolated from sun-dried apricot samples. The obtained DNA samples were then dried, and structural analyses were performed using FTIR analysis. Thermal properties were determined using thermogravimetric analysis, differential thermal analysis, and differential scanning calorimetry. The electrochemical properties of the isolated DNA structures were investigated. Finally, solutions of calf thymus DNA (ctDNA) at 2.16 ppm, 4.23 ppm, 6.23 ppm, 8.15 ppm, and 10 ppm were added to DNA samples isolated from two different apricots. Peak currents around 280 mV and 820 mV were observed to increase with increasing concentration. As a result of the increase in these two peaks alone, it can be concluded that ctDNA has two electrochemical oxidation peaks at these potentials under these optimized conditions. Since the currents of the peaks observed at other potentials remain almost constant, it can be concluded that they represent other electroactive species. Consequently, the voltammetric DNA analysis method was determined to be promising for the practical, time-saving, and low-cost identification of different apricot species.
