ENVESOME – Chemical-analysis protocols for the targeted determination and untargeted identification of priority contaminants in biological samples

This document presents the chemical-analysis protocols and human biomonitoring (HBM) analytical capacities developed and compiled within the ENVESOME project (The Environmental Exposome and Health), funded by the European Union’s Horizon Europe Research and Innovation Programme under Grant Agreement No. 101157269. The resource provides an overview of targeted and untargeted analytical methodologies available across ENVESOME consortium laboratories for the determination and identification of priority environmental contaminants and exposure biomarkers in biological samples. The analytical coverage includes persistent and non-persistent environmental chemicals such as polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), per- and polyfluoroalkyl substances (PFAS), phthalates and alternative plasticizers, bisphenols, pesticides, metals and metalloids, and biomarkers of nicotine exposure. The document describes analytical workflows applied to multiple biological matrices, including urine, blood, serum/plasma, and hair, using techniques such as LC–MS/MS, online solid-phase extraction LC–MS/MS, high-resolution mass spectrometry, gas chromatography–mass spectrometry, ICP-MS, and atomic absorption spectrometry. Both targeted biomonitoring approaches and untargeted/suspect-screening workflows are represented. In addition, the document provides an overview of human biomonitoring datasets available within the ENVESOME framework, including their populations, biological matrices, chemical coverage, and complementary health and environmental information. These datasets support exposure characterization, identification of co-exposure patterns and exposure signatures, network-based analyses, life-course exposure assessment, and the prioritization of environmentally relevant chemical mixtures. The protocols and resources compiled here contribute to the harmonized exposome-oriented analytical framework of ENVESOME and support the integration of chemical exposure data across populations, biological matrices, and complementary environmental and health datasets.
