Diagnostic and prognostic value of miR-4793-3p in bladder cancer and its regulatory role

Bladder cancer (BC) is a prevalent and aggressive malignancy worldwide, yet the role of miR-4793-3p in its pathogenesis remains poorly defined. This study aims to explore the clinical significance and potential mechanism of miR-4793-3p in BC. Serum samples from 133 BC patients and 120 healthy individuals, along with paired tumor and adjacent normal tissues from BC patients, were collected. The clinical value of miR-4793-3p was evaluated for diagnosis (ROC curve), tumor progression (correlation with clinicopathological features) and prognosis (Kaplan-Meier curve and Cox regression). Effects of miR-4793-3p on the proliferation and invasion abilities of human BC cell lines T24 and UM-UC-3 were detected through CCK-8 and Transwell assays. Downstream targets were identified via bioinformatics and dual-luciferase assays, with rescue experiments to assess TP53INP1 involvement. miR-4793-3p was upregulated in BC serum and tissues compared with healthy individuals. It showed potential to distinguish BC patients and predicted the 5-year survival. Highly expressed miR-4793-3p was associated with high-grade, advanced T stage and lymph node metastasis (LNM), which were independent risk factors for 5-year survival in BC patients. In T24 and UM-UC-3 cells, miR-4793-3p was upregulated, and its knockdown inhibited cell proliferation and invasion. TP53INP1 was negatively regulated by miR-4793-3p. Knockdown of TP53INP1 reversed the inhibitory effect of miR-4793-3p silencing on the proliferation and invasion of T24 and UM-UC-3 cells. miR-4793-3p may serve as a potential complementary biomarker for the diagnosis and prognosis of BC. Furthermore, it promotes BC cell proliferation and invasion by directly targeting TP53INP1.
