CLEANING VALIDATION IN THE PHARMACEUTICAL INDUSTRY: PRINCIPLES, REGULATORY REQUIREMENTS, CURRENT PRACTICES, AND RECENT ADVANCES – A REVIEW

Cleaning validation is a critical component of pharmaceutical quality assurance that ensures manufacturing equipment is consistently cleaned to predetermined acceptable levels before its reuse. In pharmaceutical manufacturing, the same equipment is frequently used for producing different products, increasing the possibility of cross-contamination, carryover of active pharmaceutical ingredients (APIs), excipients, cleaning agents, lubricants, and microbial contaminants. Such contamination may adversely affect product quality, efficacy, safety, and patient health. Therefore, regulatory authorities require scientifically documented evidence demonstrating that cleaning procedures effectively remove contaminants and maintain equipment cleanliness.[1-6] This review provides a comprehensive overview of cleaning validation, including its principles, objectives, regulatory requirements, validation lifecycle, sampling techniques, analytical methods, acceptance criteria, documentation practices, risk assessment strategies, and revalidation requirements. The review discusses recommendations from major regulatory agencies, including the US Food and Drug Administration (FDA), European Medicines Agency (EMA), World Health Organization (WHO), Pharmaceutical Inspection Co-operation Scheme (PIC/S), International Council for Harmonisation (ICH), and Indian Schedule M guidelines. Recent developments such as automated Cleaning-in-Place (CIP) systems, Process Analytical Technology (PAT), digital documentation, Health-Based Exposure Limits (HBEL), artificial intelligence (AI), and risk-based cleaning validation approaches are also highlighted.[2-8] Effective cleaning validation minimizes contamination risks, enhances regulatory compliance, improves manufacturing efficiency, reduces product recalls, and ensures patient safety. Continuous improvement, scientific risk assessment, and modern analytical technologies are expected to further strengthen cleaning validation practices in the pharmaceutical industry.[9-13]
