CAR T cells against the B cell receptor to precisely eliminate pathogenic B cells
Chimeric antigen receptor (CAR) T cells, monoclonal antibodies, and bispecific antibodies (BsAbs) have transformed the treatment of B cell malignancies and are emerging as an exciting therapeutic option for autoimmune diseases. In particular, CAR T cells targeting CD19 (CART19) and BsAbs targeting CD3/CD19 and CD3/CD20 are now approved for B cell malignancies and are undergoing clinical trials for B cell-mediated autoimmune diseases. However, while potent, these therapies target pan-B cell markers and are, therefore, associated with a significant B cell aplasia and hypogammaglobulinemia.
