Design of Chitosan–Gelatin Hydrogel System Infused With Moxifloxacin for Wound Healing
ABSTRACT This study aimed to develop and evaluate moxifloxacin‐infused chitosan–gelatin hydrogel (MCGH) for wound healing, focusing on optimizing drug delivery, antibacterial activity, and healing potential. Hydrogels were prepared using various chitosan‐to‐gelatin ratios (1:1, 1:2, 1:3, 3:1, and 2:1) via solvent casting method (F1–F5). Various key parameters of hydrogel batches were assessed and compared. Among these, Batch F1 containing a 1:1 proportion of chitosan‐to‐gelatin was identified as optimal, exhibiting superior characteristics as compared to other formulations. F1 revealed excellent mucoadhesive strength (15 ± 1.24 g), tensile strength (1.569 ± 0.085 kPa), and compressibility (197.8 ± 3.26 kPa). FTIR, DSC, and SEM analyses confirmed the hydrogel's structural integrity and thermal stability. F1 exhibited significant antibacterial activity against Staphylococcus aureus and Escherichia coli as compared to moxifloxacin suspension and blank hydrogel. Further, in vivo studies supported its efficacy in promoting wound healing. Furthermore, its higher water retention (19.8% ± 0.43% at 25°C), protein absorption (50.60% ± 2.43%), drug content (82.62% ± 2.93%), prolonged drug release (76.19% ± 1.42% over 48 h), strong adhesion (468.15 ± 2.53 N/m 2 ), and antibacterial properties offer both protection against infection and a conducive environment for tissue repair, making it a promising candidate for advanced topical wound therapy. Overall, moxifloxacin‐infused chitosan/gelatin hydrogel (F1) presents an effective wound care solution.
