A scalable recombinant pipeline for disulphide-stapled peptides — code and data
Analysis code and sequence-free source data for a 96-well recombinant pipeline for disulphide-stapled cyclic peptides: automated IVA primer design, qPCR cloning QC, periplasmic secretion and DOC lysis, IMAC, SEC yield analysis with hierarchical clustering, and Ellman's free-thiol staple QC. Includes the designed expression construct (His6-SUMO-linker-ccdB in pET-24b(+)). Notebooks 1, 2 and 4 run end-to-end on a bundled synthetic example; notebook 3 (SEC) reproduces Figure 4 using the third-party SAPP_DMX engine (github.com/bwicky/SAPP_DMX) plus user-supplied raw HPLC traces. Designed peptide sequences and SPR binding data are withheld (intellectual property / reported separately). Licensing: code is MIT; data and construct are CC BY 4.0.
