Abstract Introduction In a typical flow cytometer, the signal generated by a single cell crossing the focused light beam produces an electrical voltage pulse. Three meta-parameters–height (H), area (A), and width (W) of the pulse–are traditionally used to characterize cells across scatter and fluorescence channels. However, cell clumping and closely spaced events can produce overlapping pulses, leading to coincidence effects that compromise data accuracy Methods Standard doublet discrimination using area-versus-height plots often relies on subjective user judgment, underscoring the need for more robust and objective gating strategies. Results Standard doublet discrimination using area-versus-height plots often relies on subjective user judgment, underscoring the need for more robust and objective gating strategies. Conclusion Results are shown using biological samples and NIST-traceable calibration beads. Funding Source n/a Topic Categories Technological Innovations in Immunology (TECH)
