An Iodine‐Mediated [3+2+1] Povarov Strategy for Accessing Pyridocoumarins With Tunable Solvatochromic Properties and Serum Albumin‐Binding Ability
Being a fused system having both coumarin and quinoline moieties, pyridocoumarins exhibit various biological activities such as antiallergic, antidiabetic, analgesic, anti‐inflammatory and antimicrobial, etc. Three component Povarov type [3 + 2 + 1] cycloaddition reaction between acetophenones, 6‐aminocoumarin and styrenes has been developed under I 2 ‐DMSO condition to synthesize a series of 2‐benzyol‐4‐phenyl substituted fused pyridocoumarins. The reaction mechanism proceeds via Kornblum oxidation of acetophenone to result phenylglyoxal, which later undergoes Povarov type of reaction to yield the desired product. Fluorescence solvatochromic response of representative synthesized system and their interaction behavior with bovine serum albumin has also been investigated to highlight the photochemical and biomedical application potency of this class of compounds.
