Effect of oral antioxidant supplementation containing resveratrol, grape seed extract, L-carnosine, and Curcuma longa on clinical signs of skin aging: A prospective pilot study

Objectives: The objective of the study is to evaluate the clinical efficacy of a 12-week oral supplementation containing resveratrol (50 mg), grape seed extract (50 mg), L-carnosine (50 mg), and Curcuma longa extract (25 mg) on visible signs of skin aging in healthy adults. Material and Methods: This single-center, prospective, single-blinded pilot study enrolled ten adults (aged 30–52 years) seeking anti-aging treatment. Participants consumed the oral formulation daily for 12 weeks. Assessments included 3D digital wrinkle analysis using ANTERA 3D ® , clinical evaluation of sagging, and skin radiance scoring using the color, luminosity, brightness, transparency (CLBT) and Imperfections Scales. Evaluations were performed at baseline, after 12 weeks, and 4 weeks post-discontinuation. Results: Significant reductions were observed in wrinkle parameters at week 12. Small wrinkle size and width decreased significantly ( p < 0.001 and p = 0.01), while depth reduction was not significant ( p = 0.23). Medium and large wrinkles showed significant improvements across size, depth, and width ( p ≤ 0.01). Skin sagging showed a non-significant reduction ( p = 0.06). CLBT parameters demonstrated directional improvement but were not statistically significant ( p > 0.05). Conclusion: The oral antioxidant and antiglycation formulation demonstrated significant improvement in objective wrinkle parameters over 12 weeks, particularly in medium and large wrinkles. Changes in skin quality parameters were not statistically significant, likely due to the small sample size. Larger controlled studies are needed.
