Sequence-constrained revision of a sequential S8/PtpA hypothesis for Ara h 2 DPYSPS motifs
Version 2 correction. The Q69/QDP arm proposed in version 1 interrupts only the middle of three canonical Ara h 2 DPYSPS motifs, while the Q76/QDP arm interrupts none. A corrected, bounded sequence-level analysis identifies R62, Q69, and R81 as a clean three-operation candidate set, predicting release of RDP, QDP, and RDP and downstream starts Y65, Y72, and Y84. The release provides a revised preprint, a public staged validation outline, a claim ledger, and a candidate table. No biochemical experiment has been executed. No candidate enzyme activity, intact-protein three-motif cleavage, reduced IgE binding, reduced basophil activation, treatment, prevention, clinical benefit, safety, or dosing has been demonstrated. Published evidence that cleaved Ara h 2 proteoforms can retain comparable IgE binding makes a separate allergy-function gate mandatory. Proprietary computational implementation, orchestration logic, certification methods, source code, private manifests, randomization seeds, per-well layouts, decode maps, internal receipts, paths, endpoints, and participant-level data are intentionally excluded.
