AI-Driven Multidimensional Phenotyping for Variant Classification in Rare Neurogenetic Disorders
Can AI-assisted phenotyping not only improve diagnostic accuracy but also drive genetic variant classification in neurologic rare disease diagnostics? In recent years, advances in digital phenotyping have introduced computational tools that transform clinical features into quantifiable data.Deep learning-based facial analysis platforms such as GestaltMatcher analyze facial morphology and compute similarity scores between an individual patient and reference cohorts with genetically defined syndromes. 1ols such as Prioritization of Exome Data by Image Analysis extend this approach by integrating multiple phenotypic similarity metrics such as Human Phenotype Ontology (HPO)encoded clinical features, image-derived facial phenotypic scores, and exome sequencing data to rank candidate variants according to a multimodal framework.
