An Autonomous Multi-Stimuli Responsive Liposomal Matrix Conjugated with WGA Lectin for Direct Mechanical Cytolysis and Chemical Apoptosis of Hypersialylated Tumors
This framework introduces Project ADV-NANO-ONCO-2026, an innovative, cell-free, non-viral therapeutic paradigm designed to overcome the classical bottlenecks of oncological immunotherapy—specifically tumor hypersialylation and MHC-I down-regulation. The system utilizes a hybrid, nano-engineered liposomal matrix functionalized with Wheat Germ Agglutinin (WGA) lectin. By shifting the targeting strategy from standard protein receptors to glycan-carbohydrate coupling, the vesicle exploits the tumor's protective sialic acid shield as a high-affinity docking matrix, triggering automated receptor-mediated endocytosis. Internally, the vesicle incorporates a multi-tiered molecular fuse sensitive to intra-tumoral metabolic hallmarks: endosomal acidification (pH 5.0 - 4.5), high intracellular Glutathione (GSH) concentrations (10 mM), and overexpressed Cathepsin B enzymes. Upon internalization, these parallel triggers induce immediate disintegration of the lipid bilayer, releasing a dual payload: a salt-free hypotonic hydro-core that drives rapid osmotic swelling and mechanical membrane rupture (Osmotic Lysis), alongside recombinant Granzyme B proteases that independently orchestrate intrinsic molecular apoptosis. This synthetic non-viral approach completely bypasses the delays, replication risks, and plasmid dilution limitations identified in predecessor prokaryotic viral frameworks such as Project SUB-VIRO-ONCO-2026.
