Conductive Scaffold for Spinal Cord Regeneration Bridge (CSRB): A Four-Component Fiber System with Dual Inhibition Release, Topographic Axon Guidance, and Remyelination for Complete and Incomplete Spinal Cord Injury

Approximately 250–500 million people worldwide live with spinal cord injury (SCI). No FDA-approved therapy restores function after chronic complete SCI as of 2026. This paper presents the Conductive Scaffold for Spinal Cord Regeneration Bridge (CSRB): a four-component bioconductive fiber jungle gym that bridges the injury gap, simultaneously dissolves inhibitory barriers, guides motor and sensory axons into topographically correct pathways, and induces remyelination — all from a single implantable scaffold. The design integrates: (1) aligned PEDOT:PSS nanofibers (200–500 nm diameter) for axon contact guidance and electrical stimulation; (2) dual sustained-release system: ChABC (chondroitinase ABC) to dissolve CSPG scar barrier + NEP1-40 (Nogo receptor antagonist peptide) to block myelin-derived inhibition; (3) spatially segregated neurotrophin gradients — BDNF for corticospinal tract (motor),

Abstract

Approximately 250–500 million people worldwide live with spinal cord injury (SCI). No FDA-approved therapy restores function after chronic complete SCI as of 2026. This paper presents the Conductive Scaffold for Spinal Cord Regeneration Bridge (CSRB): a four-component bioconductive fiber jungle gym that bridges the injury gap, simultaneously dissolves inhibitory barriers, guides motor and sensory axons into topographically correct pathways, and induces remyelination — all from a single implantable scaffold. The design integrates: (1) aligned PEDOT:PSS nanofibers (200–500 nm diameter) for axon contact guidance and electrical stimulation; (2) dual sustained-release system: ChABC (chondroitinase ABC) to dissolve CSPG scar barrier + NEP1-40 (Nogo receptor antagonist peptide) to block myelin-derived inhibition; (3) spatially segregated neurotrophin gradients — BDNF for corticospinal tract (motor),
